Antibody Pairs

Antibody pair is two different antibodies developed against the same target that with different binding regions or epitopes. Differential epitope recognition renders simultaneous binding of both antibodies to the same target, to form sandwich ELISA format. Principle of Antibody Pair

ELISA is designed to measure the amount of the target bound between a matched antibody pair. Firstly, a target-specific antibody (capture antibody) is coated to the bottom of the wells of a microplate, which is an overnight process. Then samples, standards, or controls are added into these wells and bind to the immobilized (capture) antibody. Thus, a sandwich is formed by the addition of the second antibody (detection antibody), and a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. Finally, the intensity of this signal is directly proportional to the concentration of target present in the original specimen.
