How specific is this antibody to Adenovirus serotype 2? Will it react with the highly similar Ad5?

This antibody was generated using recombinant Ad2 hexon protein and is highly specific for Adenovirus serotype 2. While Ad2 and Ad5 belong to the same species (Human adenovirus C) and their hexon proteins are similar, this antibody has been validated to specifically recognize Ad2 without significant cross-reactivity to Ad5 in standard applications like ELISA and IF.

What fixation method do you recommend for immunofluorescence (IF) to detect the Ad2 hexon protein?

For optimal IF results, we recommend fixing your Ad2-infected cells with 4% paraformaldehyde (PFA) in PBS for 15-20 minutes at room temperature. Because the hexon protein accumulates in the nucleus during late-stage infection, a permeabilization step using a detergent like 0.25% Triton X-100 is essential to allow the antibody access to its target.
