Is this antibody specific for AES (TLE5), or does it cross-react with other Groucho/TLE family members (TLE1-4)?

The immunogen for this antibody is the N-terminal region of AES. This region contains the highly conserved Q domain, which is shared among all TLE family members. Therefore, it is possible that this antibody may cross-react with other TLE proteins (TLE1, TLE2, TLE3, TLE4). We strongly recommend validating specificity in your system, for example, by using cell lines with known expression profiles of different TLE members or by using knockdown controls.

What subcellular localization should be expected for AES?

AES/TLE5 is a transcriptional co-repressor. While it lacks a classic nuclear localization signal, it is predominantly found in the nucleus, where it interacts with transcription factors on the DNA. It can also be found in the cytoplasm. If you were to validate this antibody for immunofluorescence, you would expect to see both nuclear and cytoplasmic staining.
