What is the species reactivity?

Acetylation of α-tubulin at Lysine 40 is a highly conserved post-translational modification. This antibody has been validated for reactivity with Human, Mouse, and Rat. Based on the sequence homology of the epitope, it is expected to react with a broad range of other species, including but not limited to bovine, canine, chicken, and zebrafish. However, we always recommend validating performance in any new species.

Is it necessary to treat cells to see a signal?

This antibody is sensitive enough to detect endogenous levels of acetylated tubulin in many cell types. However, the signal can be significantly enhanced by treating cells with microtubule-stabilizing agents (e.g., paclitaxel) or HDAC inhibitors (e.g., Trichostatin A, TSA), which increase the pool of acetylated microtubules. This is a common practice when studying the dynamics of this modification.
