Is this antibody suitable for identifying new acetylated proteins?

Yes, this is an excellent tool for discovery proteomics. The recommended workflow is to perform an Immunoprecipitation (IP) using this antibody to enrich all acetylated proteins from your sample. The enriched proteins can then be identified and quantified using mass spectrometry (LC-MS/MS).

The datasheet says the immunogen was a histone H3 peptide. Why is it sold as a pan-acetyl-lysine antibody?

While the immunogen used was a specific acetylated peptide (from histone H3), the resulting antibody clone was selected for its ability to broadly recognize the acetyl-lysine modification in a context-independent manner. This screening process ensures that the antibody is not limited to its original immunogen but can bind effectively to acetylated lysine on a wide variety of different proteins.

Introduction

Acetyllysine is an acetyl-derivative of the amino acid lysine. In proteins, the acetylation of lysine residues is an important mechanism of epigenetics. It functions by regulating the binding of histones to DNA in nucleosomes and thereby controlling the expression of genes on that DNA. Non-histone proteins are acetylated as well. Unlike the functionally similar methyllysine, acetyllysine does not carry a positive charge on its side chain.
