AKAP5 (known as AKAP79 in humans and AKAP150 in rodents) is a master scaffolding protein that is highly enriched in neurons. Its primary function is to create a "signaling hub" by anchoring key enzymes to the postsynaptic density (PSD). It simultaneously binds: Protein Kinase A (PKA) Protein Kinase C (PKC) Calcineurin (PP2B) (a phosphatase) By holding these opposing enzymes (kinases and a phosphatase) together, it allows for the precise, rapid, and localized regulation of ion channels and glutamate receptors (like AMPA and NMDA receptors), which is essential for synaptic plasticity (e.g., learning and memory).

This antibody is a recombinant monoclonal (clone 22) designed for high specificity against AKAP5. While the AKAP family is large, they are structurally diverse. Function

Multivalent scaffold protein that anchors the cAMP-dependent protein kinase/PKA to cytoskeletal and/or organelle-associated proteins, targeting the signal carried by cAMP to specific intracellular effectors (PubMed:1512224). Association with the beta2-adrenergic receptor (beta2-AR) not only regulates beta2-AR signaling pathway, but also the activation by PKA by switching off the beta2-AR signaling cascade. Plays a role in long term synaptic potentiation by regulating protein trafficking from the dendritic recycling endosomes to the plasma membrane and controlling both structural and functional plasticity at excitatory synapses (PubMed:25589740).
