I'm getting a weak Western Blot signal from my whole-cell lysate. How can I improve this?

Since ACOT9 is a mitochondrial matrix protein, its concentration in a whole-cell lysate can be relatively low depending on the cell type. To significantly boost your signal, we recommend preparing a mitochondria-enriched fraction from your cells or tissue. This will concentrate the ACOT9 protein and should yield a much stronger band.

What antigen retrieval method works best for IHC on cardiac tissue?

For formalin-fixed, paraffin-embedded heart tissue, we have had excellent results with heat-induced epitope retrieval (HIER). We recommend using a 10 mM Sodium Citrate buffer (pH 6.0) and heating the slides in a pressure cooker or steamer for 20 minutes. This method effectively unmasks the epitope for strong and specific staining.
