This antibody is officially validated by Creative Biolabs for Western Blotting (WB). However, as a high-quality recombinant monoclonal antibody, it has demonstrated excellent performance in other applications as well. Customer feedback has confirmed its successful use in Immunohistochemistry (IHC) on FFPE tissues and Immunofluorescence (IF). As with any antibody, protocol optimization is recommended for non-validated applications.

Has the specificity of this antibody been tested against other ADAMTS family members?

This antibody is a recombinant monoclonal, engineered for high target specificity. It was generated using a specific immunogen from human ADAMTS7. While extensive panel screening data is not publicly available, its design principle minimizes the risk of cross-reactivity with other homologous ADAMTS family members. For critical experiments, using a negative control cell line or tissue is always a good practice.

Introduction

The protein encoded by this gene is a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) family. Members of this family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The encoded preproprotein is proteolytically processed to generate the mature enzyme. This enzyme contains two C-terminal TS motifs and may regulate vascular smooth muscle cell (VSMC) migration. Mutations in this gene may be associated with susceptibility to coronary artery disease. [provided by RefSeq, Feb 2016]

Fucosylation mediates the efficient secretion of ADAMTS family members. Also can be C-glycosylated with one or two mannose molecules on tryptophan residues within the consensus sequence W-X-X-W of the TPRs. N- and C-glycosylations can also facilitate secretion. O-glycosylated proteoglycan. Contains chondroitin sulfate.
May be cleaved by a furin endopeptidase (By similarity). The precursor is sequentially processed.
