Is this antibody specific for AFAP1L2, or does it cross-react with AFAP1 or AFAP1L1?

This monoclonal antibody was generated against the full-length human AFAP1L2 protein and has been screened for high specificity. Our validation data from Western blotting and other applications indicates that it does not cross-react with the other highly homologous family members, AFAP1 and AFAP1L1. It is an excellent tool for specifically studying the function of AFAP1L2.

Do you have a recommended protocol for immunofluorescence (IF) or immunoprecipitation (IP)?

For immunofluorescence, we recommend fixation with 4% paraformaldehyde followed by permeabilization with 0.2% Triton X-100. For immunoprecipitation, standard lysis buffers like RIPA buffer are suitable. The optimal amount of antibody for IP should be determined by titration, but a good starting point is 2-5 µg of antibody per 1 mg of total protein lysate. Introduction

Actin filament associated protein 1 like 2 (AFAP1L2) is a cytosolic adaptor protein and signal transduction mediator which is highly expressed in the thyroid and spleen. AFAP1L2 regulates cell proliferation, cell survival, cell motility and gene expression. XB130 is highly similar to AFAP and is a substrate and regulator of multiple tyrosine kinase-mediated signaling.
