Does this antibody distinguish between ACO1 (cytosolic) and ACO2 (mitochondrial)?

This antibody was generated using a recombinant fragment of human ACO1. Based on sequence homology, it is not expected to cross-react with ACO2. Our validation data shows a single band at the expected molecular weight for ACO1 (~98 kDa) in whole-cell lysates, indicating high specificity for the cytosolic form.

Does the antibody recognize both the apo-form (IRP1) and the holo-form (c-aconitase) of ACO1?

Yes, this antibody recognizes the ACO1 protein regardless of whether it contains the iron-sulfur cluster. It is suitable for detecting the total ACO1 protein pool within a cell lysate, allowing you to quantify changes in total protein expression rather than just the activity state.

Function

Iron sensor. Binds a 4Fe-4S cluster and functions as aconitase when cellular iron levels are high. Functions as mRNA binding protein that regulates uptake, sequestration and utilization of iron when cellular iron levels are low.
