The full-length human AGO1 protein (also known as EIF2C1) has a calculated molecular weight of approximately ~97 kDa. You should look for a strong band at this position. Depending on your gel and sample, you may see minor variations, but this ~97 kDa band is the target.

What is the primary function and localization of AGO1?

AGO1 is a key protein in the RNA-induced silencing complex (RISC). Its primary job is to bind to microRNAs (miRNAs) and guide the RISC complex to target mRNAs, leading to translational repression or mRNA degradation. Its localization is primarily cytoplasmic, where it is often concentrated in P-bodies (processing bodies). However, a sub-population of AGO1 is also known to function in the nucleus, where it's involved in transcriptional silencing and splicing.

Function

Required for RNA-mediated gene silencing (RNAi). Binds to short RNAs such as microRNAs (miRNAs) or short interfering RNAs (siRNAs), and represses the translation of mRNAs which are complementary to them. Lacks endonuclease activity and does not appear to cleave target mRNAs. Also required for transcriptional gene silencing (TGS) of promoter regions which are complementary to bound short antigene RNAs (agRNAs).
