What is the main difference between AGO3 and other Argonaute proteins like AGO2?

All four human Argonaute proteins (AGO1-4) are central to the RNA-induced silencing complex (RISC) and bind microRNAs. The critical difference is that AGO2 is the only one with "slicer" activity, meaning it can efficiently cleave target mRNA. AGO1, AGO3, and AGO4 are thought to act primarily by repressing translation. You would use a specific AGO3 antibody, like this one, to study its unique roles, such as its recently discovered function in modulating miRNA "passenger strand" selection or its compensatory role when AGO2 is absent.

What is the expected subcellular localization for AGO3?

Like other Argonaute proteins, AGO3 is found primarily in the cytoplasm. In immunofluorescence, you will typically see it concentrated in distinct foci known as P-bodies (processing bodies), as well as in a more diffuse pattern throughout the cytoplasm. Some studies also report a population of AGO3 in the nucleus, where it may be involved in transcriptional regulation.

Required for RNA-mediated gene silencing (RNAi). Binds to short RNAs such as microRNAs (miRNAs) and represses the translation of mRNAs which are complementary to them. Proposed to be involved in stabilization of small RNA derivates (siRNA) derived from processed RNA polymerase III-transcribed Alu repeats containing a DR2 retinoic acid response element (RARE) in stem cells and in the subsequent siRNA-dependent degradation of a subset of RNA polymerase II-transcribed coding mRNAs by recruiting a mRNA decapping complex involving EDC4. Possesses RNA slicer activity but only on select RNAs bearing 5'- and 3'-flanking sequences to the region of guide-target complementarity.

Ubiquitinated on surface-exposed lysines by a SCF-like E3 ubiquitin-protein ligase complex containing ZSWIM8 during target-directed microRNA degradation (TDMD), a process that mediates degradation of microRNAs (miRNAs). Ubiquitination by the SCF-like E3 ubiquitin-protein ligase complex containing ZSWIM8 leads to its subsequent degradation, thereby exposing miRNAs for degradation. ZSWIM8 recognizes and binds AGO3 when it is engaged with a TDMD target.
