Target

Full Name
acyl-CoA synthetase short-chain family member 2
Introduction
This gene encodes a cytosolic enzyme that catalyzes the activation of acetate for use in lipid synthesis and energy generation. demonstrated that the metabolic enzyme ACSS2 directly regulates histone acetylation in neurons and spatial memory in mammals. In a neuronal cell culture model, ACSS2 increased in the nuclei of differentiating neurons and localized to upregulated neuronal genes near sites of elevated histone acetylation. A decrease in ACSS2 lowered nuclear acetyl-CoA levels, histone acetylation, and responsive expression of the cohort of neuronal genes. In adult mice, attenuation of hippocampal ACSS2 expression impaired long-term spatial memory, a cognitive process that relies on histone acetylation.

Alternative Names
Acyl-CoA Synthetase Short-Chain Family Member 2; Acetyl-Coenzyme A Synthetase 2 (ADP Forming); Acyl-Activating Enzyme; EC 6.2.1.1; ACECS; ACAS2; ACS; Cytoplasmic Acetyl-Coenzyme A Synthetase; Acetyl-CoA Synthetase; Acetate--CoA Ligase; Acetate Thiokinase; Acetate-CoA Ligase; DJ1161H23.1; EC 6.2.1; ACSA
Function
Catalyzes the synthesis of acetyl-CoA from short-chain fatty acids. Acetate is the preferred substrate. Can also utilize propionate with a much lower affinity (By similarity).
Biological Process
Acetate biosynthetic process
Acetyl-CoA biosynthetic process from acetate
Ethanol oxidation
Lipid biosynthetic process
Propionate biosynthetic process
Cellular Location
Cytosol
PTM
Reversibly acetylated at Lys-661 (By similarity). The acetyl-CoA synthase activity is inhibited by acetylation and activated by deacetylation mediated by the deacetylases SIRT1 and SIRT3 (By similarity).
